They are part of the immune system; engulfing and digesting invading bacteria. The reactive oxygen species (ROS) indicator dihydrorhodamine 123 (DHR) has been added to these cells via passive diffusion. The neutrophils produce oxygen ions and hydrogen peroxide in order to destroy the bacterial cell walls. These ROS molecules oxidize the indicator to hydrorhodamine which fluoresces yellow when stimulated with the laser light from the microscope.
