Enzymes are synthesised by ribosomes (dark blue) on the endoplasmic reticulum (left). After synthesis the hydrolase enzyme is transported to the Golgi apparatus (right) for modification. Here, it is tagged with a mannose-6-phosphate molecule (P), which marks it for transport to a lysosome (bottom). Once in the lysosome an ATP (adenosine triphosphate) driven proton pump transports protons into the lysosome. This creates an acidic environment that causes the mannose-6-phosphate to disassociate, and also activates the hydrolase. The mannose-6-phosphate molecule is carried back to the Golgi so that it can be reused.
